Abstract

This work involved firstly optimizing the protoplast yields for F. culmorum 159026, then setting up a system for DNA-mediated transformation. Higher protoplast yields and more rapid regeneration were obtained when the organic stabilizers sucrose and sorbitol were used rather than NH4Cl. Successful transformation was achieved using the plasmids pAN7-1 and pHRC containing the E. coli hygromycin B resistance gene (hygromycin B phosphotransferase, hpt) fused to Aspergillus nidulans expression signals.
